The PI3K/AKT/mTOR pathway is a central intracellular signaling cascade that regulates cell growth, proliferation, metabolism, and survival. Activation begins when receptor tyrosine kinases or G protein-coupled receptors stimulate phosphoinositide 3-kinase (PI3K), which phosphorylates PIP2 to generate PIP3 at the plasma membrane. PIP3 then recruits AKT (protein kinase B) and PDK1, leading to AKT activation through phosphorylation.
Activated AKT phosphorylates numerous downstream substrates that control diverse cellular processes. Among its most important targets is the mechanistic target of rapamycin (mTOR), which exists in two distinct complexes: mTORC1, a master regulator of protein synthesis and autophagy, and mTORC2, which participates in cytoskeletal organization and AKT activation. Small-molecule modulators targeting specific nodes in this pathway allow researchers to dissect the contributions of individual kinases and regulatory mechanisms.
Negative regulation of the pathway is primarily mediated by the lipid phosphatase PTEN, which dephosphorylates PIP3, and by TSC1/TSC2, which inhibits mTORC1.
